- W184448433 abstract "Proteomics is a systematic approach to characterize the expressed protein complement (the proteome) present in particular tissues, cells or subcellular fractions under defined conditions. This analysis is based on (a) high resolu-tion Separation of proteins (most often carried out by two-dimensional gel electrophoresis) (b) protein identification (most often carried out by mass spectrometry) and (c) bioinformatics (evaluation of gel images, mass spectra and genome data bank searches). The protein complement of purified plant mitochondria have been displayed by gel electrophoresis and individual proteins identified by various techniques for many years. These studies have given insights into the plasticity of the mitochondrial proteome in response to environmental stress and genetic alteration. The relative contributions of the organelle and nuclear genomes to the mitochondrial proteome have been investi-gated and the intra-organellar location of particular protein components have been highlighted. Puriflcation of native protein complexes such as the electron transport complexes gave insights into the way in which these proteins operated together and identified important differences between plant and other eukaryotic mitochondria. However, despite these efforts there was still much about the mitochondrial proteome in plants that remained a mystery. Recent advances in peptide mass spectrometry coupled to the increasing sequencing of plant genomes has revolu-tionized proteome analysis. For the flrst time the rapid analysis of whole protein complements rather than just analysis of a small number of selected Polypeptides was possible. Using these techniques, mapping studies have begun to uncover the full extent of the complex protein mixtures that make up plant mitochondria. These studies are directly linking proteins to particular members of gene families, revealing proteins of unknown function and identifying novel and/or poorly investigated aspects of mitochondrial function in plants." @default.
