The only radioactive compound that is considered is tritiated thymine or its derivative thymidine. To use thymine as a precursor of DNA, it is essential to have an organism that can incorporate this compound from the medium and utilize it without degradation. These conditions are most easily met by using thymine-requiring strains. In every case, it is advisable, when working with new strains, to compare radioactive samples that have been treated with DNase with those that have not to be sure that the label has gone uniquely into DNA. Once the labeled cells are obtained, the only equipment necessary is microscope slides, a dark room, and a good phase contrast microscope. The preparation of slides and application of film are discussed in the chapter. The technique that can be very useful is to allow labeled bacteria to grow into small colonies, which are then treated for autoradiography. In this case, cells are labeled as desired, washed free of radioactive medium, and allowed growing into microcolonies of about 16–32 cells on agar-coated slides. These slides should be examined from time to time to determine the rate of growth, and the slides should not so moist that bacteria will swim free and start new colonies that are not directly derived from the cells of the original culture." @default.
