- W185023849 abstract "Publisher Summary This chapter focuses on the neurotrophic factors for experimental treatment of motoneuron disease. Experiments designed to elucidate the pharmacokinetics, binding proteins, and availability of these factors for motoneurons are necessary in order to improve strategies for planning clinical trials with such factors, and thus to provide a basis for evaluating the potential usefulness of these trophic factors for the treatment of human disease. Motoneurons express insulin-like growth factor (IGF)-receptors, and it has been observed that the expression of IGFR is increased in motoneurons of patients suffering from amyotrophic lateral sclerosis. These data suggest that indeed IGF could play a role in the maintenance of motoneurons during postnatal life. Experimental treatment of patients with motoneuron disease has shown that subcutaneous injection of IGF has beneficial effects. Insulin-like growth factor-I (IGF-I) has been shown to be a potent survival factor for embryonic chicken and rat motoneurons. At maximal concentrations, its potency in serum-free cultures of rat motoneurons is similar to that of brain-derived neurotrophic factor (BDNF). Interestingly, although IGF-I is also active on lesioned facial motoneurons in newborn rats, the effects are significantly lower than those observed with CNTF or BDNF. It could be that this is due to the presence of inhibitory IGF binding proteins in nerve tissue." @default.
