- W187773 abstract "This chapter discusses RNA–DNA hybridization experiments. The measurement of RNA–DNA hybridization is a direct approach that may eventually establish the number of V genes in the germ line. It is apparent from the range of antibody specificities that the number of different V genes expressed in a mature animal is very large. A minimum value would probably be several thousand and the actual number may be much higher. If the germ line theory is correct, all these genes would be present in every diploid cell. It is theoretically possible to estimate the number of genes by RNA–DNA hybridization. By culturing the tumor cells in a radioactive medium or by trace-labeling with 125I after isolation the mRNA can be obtained with a specific radioactivity high enough for hybridization experiments. Purification is facilitated by the fact that a large fraction of the mRNA in a typical mature plasma cell (∼ 30%) is translated into immunoglobulin." @default.
