- W1884700913 abstract "Mammalian drug-metabolizing enzymes (DMEs) are a group of cellular catalysts responsible for the metabolism of all xenobiotics and certain endobiotics in the body. Fittingly, DMEs are exceedingly versatile enzymatic systems. DMEs catalyze oxidation, reduction, conjugation, and nucleophilic trapping reactions. Unlike classical, well-behaved enzymes, DMEs exhibit common properties characteristic of promiscuous catalysts for substrate binding, catalysis, reaction type, metabolite formation, and enzyme kinetics. At the molecular level, DMEs appear to have used all means of chemical–protein interactions that are also observed in other biological systems, only that they are pushed to the extreme to achieve enzymatic versatility at the expense of substrate specificity and catalytic efficiency in DME reactions. DME promiscuity has implications in the evaluation and prediction of the safety, drug–drug interaction, and pharmacokinetic properties of therapeutic agents, as well as the risk assessment of environmental carcinogens and toxicants.Keywords:drug-metabolizing enzymes;cytochrome P450;promiscuous enzymes;drug metabolism reactions;catalysis;structures" @default.
