- W1887184089 abstract "El objetivo de este estudio fue evaluar la eficacia de las tecnicas de citometria de flujo y analisis citogeneticos en la determinacion de los niveles de ploidia de plantas de arroz (Oryza sativa L.) regeneradas a partir de las microsporas de anteras, empleando el contenido de ADN nuclear y el numero de cromosomas de plantas verdes y albinas regeneradas. Se emplearon plantas haploides (x), dobles haploides (2x) y triploides (3x) de los cultivares ‘Fonaiap 2000’ y ‘Blue Belle’, con un contenido promedio de 0,47; 0,95 y 1,38 pg de ADN, respectivamente. El nivel de ploidia de las plantas regeneradas se corroboro a traves de los analisis citogeneticos. No se encontro variacion cromosomica entre las plantas verdes y albinas regeneradas, a pesar que se ha reportado que las plantas verdes tienden a poseer menor variacion cromosomica. Las metodologias empleadas constituyeron un metodo rapido para la determinacion de los niveles de ploidia de las plantas de arroz regeneradas. Los conocimientos acerca del contenido de ADN y numero cromosomico de las plantas evaluadas proveen informacion relevante para los mejoradores y geneticistas interesados en emplear el cultivo de anteras en los programas de mejora del cultivo. Evaluation of two methodologies to determine ploidy levels in rice (Oryza sativa L.) plants regenerated by anther culture ABSTRACT The efficiency of flow cytometry and citogenetic technique were evaluated to determine the ploidy levels in rice (Oryza sativa L.) plants regenerated from microspores-anther. The nuclear DNA content and chromosomal numbers of green and albino plants regenerated were used. Haploid (x), doubled-haploid (2x), and triploid (3x) plants from ‘Fonaiap 2000’ y ‘Blue Belle’ cultivars were used, containing an average 0.47, 0.95, and 1.38 pg of DNA, respectively. The ploidy level of regenerated plants were corroborated through cytogenetic analyses. In terms of cytogenetic stability, green plants had less chromosomal variation than albino plants; however, no chromosomal variation was found. The procedures are suitable for rapid determination of the ploidy levels of rice microspore-derived plants. The DNA content and chromosomes number of plants obtained provide useful information to plant breeders and geneticists interested in using anther culture in their breeding programs.
