- W19005690 abstract "The mechanisms linking obesity to its metabolic complications are extremely complex and remain hotly debated. It is hypothesized that reasons for variability in susceptibility to obesity-mediated metabolic complications involve both environmental and genetic factors. While advances in the knowledge of variation in the human genome have led to the identification of susceptibility genes contributing to obesity and related disorders, relatively few studies have focused specifically on interactions between obesity and genetic polymorphisms in the development of metabolic complications. With genome-wide association studies (GWAS) and more comprehensive candidate gene studies, there have been tremendous advances in the identification of common gene variants that have main effects on body mass index (BMI) (and other adiposity measures), lipids, and blood pressure. However, there have been relatively few investigations that have evaluated the interactions between genetic variation and obesity on the development of metabolic syndrome traits. A common theme for common variants for complex diseases and traits, including metabolic syndrome traits, is that individual susceptibility loci have very small main effect sizes. However, it is still possible that some of these loci have larger effects on metabolic phenotypes in the presence of obesity or when provoked by a certain dietary or other lifestyle factor. Identification of these functional variants will provide additional insight into disease pathophysiology and may provide the scientific rationale for the design of more individualized prevention and treatment strategies for obesity and its metabolic complications." @default.
