Trichogrammatoidea lutea and Trichogrammatoidea cryptophlebiae are indigenous to South Africa and have considerable potential as biological control agents of various economically important agricultural pests in the country. Unfortunately, rapid, accurate means of identifying these species are not available. DNA sequence analysis of the internal transcribed spacer 2 (ITS 2) region of the ribosomal gene complex has previously been applied for identification and distinction of Trichogramma species, but not for members of the closely-related genus Trichogrammatoidea. In this study a protocol was developed that allows DNA sequence determination of the above region, following genomic DNA extraction from a single Trichogrammatoidea wasp. DNA sequence analysis of the ITS 2 region revealed both length and within-sequence variation, indicating that T. lutea and T. cryptophlebiae are distinct species. This information can be used for simple and rapid identification of the two species based on a PCR reaction of extracted DNA, avoiding time-consuming slide-mounting procedures and the availability of expert knowledge for morphological identification." @default.
