• W193148934 abstract "歯周病関連細菌のひとつであるPorphyomonas gingivalis (P.g) のextracellular vesicles (ECV) が, 好中球の各種機能を発現させるために必要な基本的因子である細胞形質膜流動性に与える影響を検討した。P.g ATCC 33277から硫安塩析法にCECVを調製し, 透過型電子顕微鏡観察, 電気泳動, および化学組成分析を行うことによってECVが分離調製されたことを確認した。好中球は全身的に健康でかつ歯周疾患の認められない成人の末梢血から分離した。調製されたECVを各種濃度 (0, 10, 50, 100μg/ml) で好中球に作用させた後, 5-あるいは16-stearic acidラベル剤 (5-SAL, 16-SAL) を好中球に取り込ませ, 電子スピン共鳴装置を用いたスピンラベル法にて膜流動性の測定とcytochrome C還元法による活性酸素産生量の測定を行つた。その結果, 好中球形質膜表層 (5-SAL) で膜流動性を有意 (p<0.01) に低下させた。しかし, ECVは各濃度において形質膜深層 (16-SAL) では膜流動性に有意な影響を及ぼさなかった。さらに, ECV (100μg/ml) はN-formyl-methionyl-leucyl-phenylalanine, phorbol myristate acetate刺激下で好中球の活性酸素産生を有意p<0.01, p<0.05) に抑制した。以上のことから, 歯肉溝あるいは歯周ポケットや歯肉組織中に遊走した好中 (球は歯周病関連細菌由来ECVによって影響を受け, 膜流動性および細胞機能が低下する可能性が示唆された" @default.
