- W1937277105 abstract "This chapter discusses a novel gene in the Armadillo family that interacts with Presenilin 1 (PS1). PS1 is the most commonly mutated gene in early-onset Alzheimer's disease (AD). Armadillo and its vertebrate homolog, β-catenin, lie downstream in the Wg/Wnt pathway. Mutations in either Armadillo or Wg cause similar phenotypic changes in segment polarity when mutated in Drosophila , and are required for the correct expression of a large set of genes including engrailed in the ectoderm and other homeobox genes in the endoderm and mesoderm. The demonstration of an interaction among PS1 and at least two members of the Armadillo family, β-catenin and δ-catenin, raises the possibility that the interaction is regulated by signals known to affect β-catenin stability, nuclear translocation, and ultimately gene transcription. The upstream signals known to regulate this pathway are probably initiated by binding of the Wg/Wnt ligand to the frizzled receptor. It is suggested that although PS1 is thought to be resident in the endoplasmic reticulum, Wnt, as a secreted glycoprotein, probably lies in an export pathway." @default.
