The protozoa, Tetrahymena pyriformis has been shown to be capable of the rapid degradation of the spingolipid bases by reactions analogous to those found in higher organisms. 2. Cell-free preparations from lyophilized cells retain the ability to phosphorylate the sphingolipid bases, but the activity for cleaving the phosphorylated long-chain bases is almost completely lost. The characteristics of the kinase reaction have been investigated using these lyophilized preparations. 3. The sphingolipid base was not phosphorylated when the amino function was blocked by acyl groups, and analogs of the long-chain bases, containing a phenyl group at carbon 3 rather than an alkyl group did not function as substrates. 4. In addition to the sphingolipid base, the kinase required Mg2+ or Mn2+, and a nucleoside triphosphate. Under the conditions of this study, CTP, ATP, UTP and CTP all served as phosphoryl donors. 5. The possibility that the rapid degradation of sphingolipid bases may be a means to generate alkyl groups and ethanolamine is discussed." @default.
