- W1964021732 abstract "It is generally accepted that K+ ions have a direct stimulatory effect on corticosteroid biosynthesis. The mechanism of action remains to be clarified. First there is the question whether the cell membrane is involved. The fact that small increments of the potassium concentration of the incubation medium stimulate corticosterone and aldosterone biosynthesis by calf adrenal cortex slices, while a homogenate of this tissue is unaffected even by large increments of potassium, leads to the concept of a membrane mechanism. An observation which gives support to this postulation is the strong inhibitory effect of ouabain on corticosterone biosynthesis in calf adrenal cortex slices (Wellen & Benraad, Biochim. Biophys. Acta (Amst.) 183, 1969, p. 110; Benraad & Wellen, Excerpta Med. Int. Congr. Ser. No. 210, 1970, p. 113). Recent studies revealed that the inhibitory effect of ouabain is observed only when corticosterone biosynthesis is stimulated by K+ ions. Again no effect is observed when a homogenate is used. At low concentrations of ouabain (10-6 M), the inhibitory effect can be prevented by raising the potassium concentration of the medium. These results led us to suppose that the stimulatory action of K+ ions involves receptor sites for K+ at the outside of the cell membrane and that ouabain acts as an inhibitor by interfering with K+ at these sites." @default.
