One includes the MDR1 genes of L. donovani and L. enriettii [1-3]. Overexpression of MDR1 by either transfection or gene amplification confers resistance to hydrophobic drugs including vinblastine and puromycin. This phenotype is similar to that conferred by overexpression of mammalian MDR1 genes, where these P-glycoproteins mediate decreased drug accumulation by increasing ATP-dependent drug efflux [19,20]. Decreased puromycin accumulation was also found in Leishmania overexpressing MDR1, although the relative contribution of efflux and influx could not be determined [1].
