A response to 5 μM ionomycin is observed from about one-third of the experimental population after 30 min and from most eggs after 1.5 hr of exposure. Secretion in response to 10 μM A23187 requires about 4 hr of exposure to the ionophore. Normal cell wall formation (triggered by fertilization) is initiated at one area of the egg surface but spreads evenly, in contrast to the patchy secretion stimulated by ionophores. Patchy secretion of cell wall material also occurs 2.5 hr after eggs are placed in low-sodium (N-methylglucamine-substituted) ASW. Eggs lyse within minutes when exposed to 5 μM ionomycin in 2.5 mM Na+ (450 mM N-methylglucamine) artificial seawater (ASW). Eggs become pear-shaped at about the time fertilized controls germinate in response to treatments such as low-sodium ASW and calcium ionophores, but they do not cleave normally. Choline has side effects upon Fucus eggs and is not recommended as a sodium substitute in this system. These results suggest that Na+Ca2+ antiport might be one mechanism by which fucoid eggs regulate internal free calcium, and they offer a possible explanation for the relative insensitivity of fucoid eggs to calcium ionophores." @default.
