In the past, this method has been used both with and without compensation for differences in the osmotic pressure of the test solutions. We studied the significance of perfusate osmotic pressure of solutions between 290 and 400 mOsmols/L (approximately 1–1.4 times normal) in guinea pigs by perfusing scala tympani with artificial perilymph solutions made hypertonic with sucrose. The cochlear microphonic-potential (CM), summating-potential (SP), and action-potential (AP) responses to 7-kHz tone bursts were recorded by differential electrodes in the first turn. The solutions with osmolality 316 mOsmols/L and higher significantly altered the cochlear potentials. The normally negative SP became positive. The AP was depressed to 50% of the preperfusion amplitude for up to 8 min after perfusion. The CM was sometimes augmented after a perfusion.
