- W1966006938 abstract "We have developed a novel water soluble oxetanyl sulfoxide (MMS350) which is a DMSO analog with radiation protector and mitigator properties. We sought to demonstrate that MMS350 protects and mitigates radiation damage in vivo. Using a C57BL/6NHsd bone marrow stromal cell line, MMS350 was added before or after irradiation ranging from 0 to 8 Gy. Cells were plated in 4 well tissue culture plates, incubated 7 days at 37°C, and stained with crystal violet with colonies greater than 50 cells counted. Data was analyzed using linear quadratic or single-hit, multi-target models. C7BL/6NHsd mice were irradiated to 9.5 Gy and injected 4 or 24 hr after irradiation with 20 mg/kg MMS350 in water, and followed for development of the hematopoietic syndrome. Data was analyzed using a Log Rank Test. Stromal cells incubated with MMS350 before irradiation were radioresistant with an increased shoulder on the survival curve (n of 15.8 ± 2.9) compared to control irradiated cells (n of 5.8 ± 1.1, p = 0.0039). Cells incubated with MMS350 following irradiation were also radioresistant: increased Do of 2.4 ± 0.3 compared to 1.9 ± 0.1 Gy for the control irradiated cells (p = 0.0444). MMS350 also mitigated 9.5 Gy total body irradiation. Mice injected with MMS350 at 4 or 24 hr after 9.5 Gy had significantly increased survival (p = 0.0092, for both time points). MMS350 is a safe and highly water soluble drug that has both radiation protection and mitigation properties." @default.
