- W1966698891 abstract "Abstract Acid detergent pretreatment methods dissolve most of the hemicellulose and other fractions leaving insoluble alphacellulose, ash, and lignin. In the neutral detergent procedure, the hemicellulose is also (in theory) insoluble. These methods have both been used successfully as pretreatment methods for the determination of lignin, and both have the potential to provide values for cellulose. This study identifies and quantifies the fraction remaining after various treatments. We evaluated the neutral detergent fibre cellulose composition, and quantified the effect of including EDTA, wetting agent, and sodium sulphite. The variation in cellulose fractionation following the inclusion of alternative wetting agents was also monitored. Plant material was fractionated using the classical holo, alpha, and hemicellulose methods prior to and after the detergent methods. Cellulose levels were higher when EDTA was present in the fibre stage, supporting the theory that its presence inhibits the dissolution of hemicellulose. The 72% soluble fraction of the fibre following ADF(SLS) pretreatment was found to be correlated with the holocellulose, presenting a rapid simple method for the determination of holocellulose from the lignin method." @default.
