- W1966861750 abstract "Obesity is an emerging problem in the Western hemisphere, and is associated with increased risk for Alzheimer's disease. Despite the many medical problems associated with increased consumption of high-fat diets, mechanistic effects of the “Western” type of diet on brain morphology and function have not been revealed. The purpose of this study was to examine functional and structural correlates of continued feeding of a diet rich in saturated and hydrogenated fatty acids upon the limbic system of aged rats. Young or aged Fischer 344 rats were treated with normal rat chow supplemented with 10% hydrogenated coconut oil and 2% cholesterol for 6 months. Working and reference memory was tested using a radial arm maze, and brain tissue was examined for vascular, glial, and neuronal constitutents. Our data demonstrate that aged rats were more sentitive to long-term treatment with the high-fat diet than young rats and displayed significant reduction in memory function. Examination of BBB specific proteins as well as glial markers revealed increased inflammation and a breach of the BBB in high-fat-fed subjects. The data suggest that memory loss caused by high-fat intake may be due to altered BBB properites leading to elevated neuroinflammation in the brain." @default.
