- W1967714912 abstract "Memantine is approved in over 75 countries worldwide for the treatment of Alzheimer's disease (AD). A novel once-daily 20mg formulation has been approved in Europe. Previous placebo-controlled studies have shown positive memantine effects on cognition, activities of daily living, and behavior with a favorable safety and tolerability profile. Analyses of these studies suggest that memantine treatment may benefit language and communication abilities. This trial studied the effects of once-daily 20 mg memantine treatment on cognition and functional communication in patients with moderate to severe AD. Outpatients diagnosed with AD (N=97; 56 % women; age: 53-91 years; MMSE range: 3-19) participated in a 16-week, multi-national, single-arm, open-label study of memantine. After a 4 week titration from 5 mg to 20 mg, memantine treatment was continued at a maintenance dose of once-daily 20 mg, followed by a 4-week wash-out period. The primary endpoint was the baseline-to-12 week endpoint total score change on the Consortium to Establish a Registry for Alzheimer's disease-neuropsychological battery (CERAD-NP). A secondary endpoint was the Functional Linguistic Communication Inventory (FLCI). Analyses were based on the full-analysis set population, assessed using a one-sided t-test. Safety and tolerability were assessed by recording treatment-emergent adverse events (TEAEs). For the primary endpoint, the CERAD-NP total score improved significantly after 12 weeks of memantine treatment compared to baseline (5.9 ± 8.8 SD, n=92, p<0.0001). For the secondary endpoint, the FLCI score improved significantly after 12 weeks compared to baseline (4.4 ± 6.8, n=92, p<0.0001). All endpoints also showed significant improvement after 4 and 8 weeks of treatment and after 4 weeks of wash-out compared to baseline. At least one TEAE was reported by 38 (39%) patients. Treatment with once-daily 20 mg memantine significantly improved cognition and functional communication skills in patients with moderate to severe AD. The changes were seen as early as 4 weeks following the start of treatment and persisted after a 4 week wash-out period of memantine." @default.
