• W1968444198 abstract "<h2>Abstract</h2> A preparation rich in sarcolemma (SL) has been isolated from dog heart. Ventricular fibers, washed until uncontaminated on phase interference microscopy, were extracted with KI leaving SL enclosing residual mitochondria (M) and sarcoplasmic reticulum (SR). This SL preparation contained the Na-K stimulated, ouabain sensitive ATPase of the heart. The Na-K ATPase specific activity of SL and paired M and SR preparations were judged by Na-K-Mg ratios and ouabain sensitivity. The ratio for SL varied from 2 to 7, values comparable to those of RBC ghosts. Values for SR, M and fibers were 1.0, 0.9 and 1.2. Ouabain inhibition of SL had a threshold of 10⁻⁷ m, equaling 53% inhibition of the Na-K stimulation. At these concentrations there were no effects on SR and M. Further, the SL Na-K ATPase was unaltered by DNP, azide and oligomycin but was removed by deoxycholate and depressed by 10⁻³ m Ca. We have shown that cardiac SR has the Ca capacity and uptake rate required for physiologic relaxation. In contrast, SL had no rapid Ca uptake. This represents the initial isolation of myocardial SL, localizes the Na-K ATPase to this organelle and provides a subcellular site for control of Na movements and a site of action for digitalis." @default.
