- W1969773791 abstract "We have employed lambda phages carrying a temperature-sensitive mutation in the gam gene to study gam gene control over the host recBC DNAase and the effects of this control on lambda DNA replication in vivo . Our results show that the gam ts-mediated inhibition of the BC nuclease can be reversed by a temperature shift. Such activation of the BC nuclease at a late time after infection results in a preferential degradation of concatemers, presumably formed as tails on rolling circles. Relaxed and covalently closed circular monomers, normally formed as the products of early replication, are resistant to enzyme action. Destruction of concatemers seems to involve only the double strand-specific exonuclease activity of the BC nuclease complex. After a late temperature shift, DNA synthesis continues at a substantially higher rate than BC nuclease-mediated enzymatic digestion of concatemers. Furthermore, rolling circle replication continues to predominate, and only a limited fraction of the molecules is actually reversed to early, circular replication. We believe that this fraction may consist of transient intermediates of special structure containing free ends (sensitive to exonucleolytic attack), the formation of which provides a ratelimiting step in late DNA replication. BC nuclease action on these hypothetical intermediates would inhibit the transition from early to late lambda replication." @default.
