- W1970156426 abstract "In a previous report, we demonstrated that newly developed non-viral Aβ DNA vaccines are safe and effective in Aβ reduction in the cerebral cortex and hippocampus of Alzheimer's disease model mice (APP23 mice) (PNAS, 103, 9619-24, 2006). The present study was undertaken to examine whether DNA vaccines effectively reduce Aβ deposition in the brain of aged rhesus monkeys. DNA vaccines at a does of 3 mg were injected intramuscularly on a biweekly basis to rhesus monkeys (16-18 years old, all females, n=3). The same amount of empty vector was injected in the control group (n=3). Before and during vaccination, blood was drawn and served for hematological and biochemical examinations. The levels of cytokines and chemokines were determined using Bio-Plex ™ Suspension Array System (Bio-Rad, Japan). Six months after the first vaccination, the brains of treated and control monkeys were sampled and brain sections were immunohistochemically stained for microglia and Aβ depositions. Quantitative analysis of Aβ deposits was performed using the Image J software. DNA vaccination reduced Aβ burden to approximately 50% of that found in control monkeys. Quantitative analysis revealed a significant Aβ reduction in treated monkeys (28378 ± 14852 pixels) compared to control monkeys (59057 ± 4423 pixels, p = 0.026). Histological examinations with hematoxylin and eosin and Berlin blue staining showed neither inflammatory cell infiltration nor microhemorrhage. Examination of the heart, liver, spleen and kidney showed no pathological changes. There were no detectable levels of cytokines (IL-2, IL-4, IL-10, TNF-α and IFN-γ) and chemokines (MIP-1α, MIP-1β, MCP-1 and RANTES) in the blood throughout the observation period. One monkey in the treated group showed transient increase in leukocytes; however, it was not considered as the side effect of vaccination. DNA vaccines produced in our laboratory was effective in Aβ reduction in the brain of aged rhesus monkeys without obvious side effects such as neuroinflammation and microhemorrhage. The findings obtained in this study provide useful information in performing preclinical and clinical trials." @default.
