- W19711271 abstract "This chapter reviews immunological tolerance from the viewpoint of the insights into the phenomenon that can be gained by studying the effects of antigen dose, antigen molecular weight, and antigen fate in vivo . Several examples are discussed in which tolerance can be obtained with either high or low dosages of antigen. The chapter describes the extraordinary and poorly understood phenomenon of ultralow zone tolerance to flagellar antigens in which amounts of antigen as low as 10 -16 gm/gm body weight/day can induce significant partial tolerance. The distribution of labeled antigen molecules during the in vivo induction of tolerance fails to reveal a single common pattern that covers all experimental circumstances. In many models, wide diffusion and prolonged extracellular persistence of tolerogenic antigen is seen, making it likely that lymphoid cells are rendered tolerant by contacting free, non-processed antigen. In other circumstances, the chief concentration of antigen during tolerance induction is in the lymphoid follicles, raising the possibility that antigen, firmly attached to and concentrated at the surface of dendritic follicle cells, is responsible for tolerance induction." @default.
