In this chapter, we describe the main steps involved in the development of tissue-selective steroid receptor knockouts by Cre/loxP technology. Problems in the development of a mouse strain with a floxed receptor allele, the selection of a suitable Cre expressing mouse strain, the generation of cell-selective knockouts by crossbreeding of the mentioned mouse strains, and the control of appropriate receptor inactivation are discussed taking the generation of mice with a Sertoli cell-selective ablation of the androgen receptor as an example." @default.
