The reliability of such measurements is governed by the degree of purification achieved and the specificity of the colour reaction. When determinations are made on limited quantities of body fluids or tissues, absolute purification (isolation) is rarely possible and in many reactions a »background« colour due to impurities is superimposed on the absorption spectrum of the compound to be determined. Allen (1950) suggested the use of a correction formula which is applicable in cases where the light absorption of contaminating substances changes linearly with the wavelength. Borth (1956) has pointed out the advantages of this method over earlier approximations.
