- W1974867413 abstract "Abstract Newly synthesized mammalian cell DNA which is adsorbed to nitrocellulose at higher ionic strengths and neutral pH can be recovered for the most part by elution with a 0.5 % sodium dodecyl sulfate solution under conditions which prevent denaturation. A method is described which permits large scale isolation of this DNA species from common DNA preparations. The DNA species isolated by this method contains nicks in the newly synthesized strand but no significant amount of single stranded segments. It is virtually identical with the newly synthesized DNA which exhibits a higher affinity to methylated albumin kieselguhr than the bulk DNA of the cells. The DNA which is retained in the interphase during DNA purification procedures using organic solvents contains large amounts of this newly synthesized DNA fraction. The increased affinity to nitrocellulose and to methylated albumin kieselguhr appears not to be caused by single stranded segments in this newly synthesized DNA but rather by attachment to other cell components, probably membrane-like structures." @default.
