- W1974991843 abstract "RATIONALE: Differential expression of homing markers in cord blood (CB) and infant blood (IB) may predict allergy. We investigated whether infants, who later developed atopic eczema (AE), demonstrated different immunological markers and proliferation responses to allergens. METHODS: CB (n = 14) and IB at 1year (n = 12) was obtained from infants with a family history of atopy and grouped according to development/non-development of eczema. T-cell proliferation was determined by CFSE dilution assay. Surface markers including cutaneous lymphocyte antigen (CLA) and integrin beta-7 (β7), homing markers for skin and gut respectively, were measured using monoclonal antibodies and flow cytometry. RESULTS: Infants who developed AE (n = 6) showed a significantly greater proliferation ratio of CD4+ and CD8 + T-cell subsets after challenge with ovalbumin (OVA) or beta-lactoglobulin (BLG) than those in the non-eczematous(NE) group; OVA AE 4.6 ± 1.1 versus NE 0.7 ± 0.8, p < 0.05 and BLG AE 4.1 ± 1.1 versus NE 1.6 ± 0.5, p < 0.05. CLA expression was detected in CB and IB. Higher expression of CLA was detected in CB from the AE (5.6 ± 2.8%) than the NE (1.0 ± 0.5%) group, p < 0.05, and a similar trend towards higher expression was seen in IB. Overall, β7 expression was significantly higher in CB than in IB (p < 0.05). No difference in β7 expression in CB was detected between the AE and NE groups. No changes in expression of CLA or β7 were seen in either group upon allergen challenge of mononuclear cells from CB and IB. CONCLUSIONS: Infants who developed AE showed increased expression of CLA premorbidly. Allergen challenge induced proliferation of TC but did not upregulate homing marker expression." @default.
