The present state of knowledge about the structure of the surface carbohydrates (sphingoglycolipid, glycoprotein and free polysaccharide) is reviewed and related to the chemical structure and possible conformation of the carbohydrate components. 2. The structure of the surface coat as revealed by light and electron microscope techniques (scanning, shadowing, replication, freeze etching, thin sectioning, negative staining, etc.) is discussed. The electron microscope problem of how attempts can be made to directly resolve individual molecular assemblies of the carbohydrate components is discussed with respect to: (a) preserving the surface coat in its natural wet state, (b) obtaining adequate contrast and resolution, and (c) minimizing electron beam radiation damage. Some recent progress in instrumentation to meet this goal is reported. 3. The significance of obtaining better morphology of cell surface coats to the problems of decreased adhesiveness, greater surface mobility and loss of contact inhibition of cancer cells is briefly reviewed." @default.
