- W1976773314 abstract "Nitric oxide (NO) is involved in the regulation of vascular basal tone and blood pressure (BP). Several polymorphisms of ENOS, the gene that codes for endothelial NO synthase, have been associated with essential hypertension. The aim of the present study was to look for a potential linkage of the 3 main diallelic polymorphisms (Glu 298 Asp, intron 4 VNTR and T-786C) and the highly informative intron 13 CA-repeat of ENOS to BP as a continuous trait. In 55 dizygotic twin pairs of the same gender (median age: 46 years) drawn at random from the birth registries of the cities of Hasselt and Sint-Truiden (Belgium), clinical and ambulatory BP were determined, and genotyping was performed by RFLP (Glu 298 Asp), PCR (intron 4 VNTR), ASO (T-786C) or automated fluorescent genotyping (intron 13 CA-repeat). None of the subjects included was on antihypertensive medication. The influence of these polymorphisms on office systolic (SBP) and diastolic (DBP) blood pressure as well as day- and night-time ambulatory SBP and DBP was assessed by sib-pair analysis. The frequency of the homozygous wild-type, heterozygous and homozygous variant genotypes were (36, 50 and 14%) for Glu 298 Asp, (82, 18 and 0%) for intron 4 VNTR and (40, 42 and 18%) for T-786C, in agreement with previous studies in Caucasians. The distribution of the different alleles of intron 13 CA-repeat was also similar to that previously described. A significant effect of intron 13 CA-repeat on day-time ambulatory SBP (p=0.01) and a borderline significant effect of the Glu 298 Asp polymorphism on day-time ambulatory DBP (p=0.05) were observed. These effects were maintained after adjustment for body mass index (BMI) and urinary sodium excretion. No effect was observed for the 2 other diallelic polymorphisms. This is the first quantitative trait linkage study assessing the effect of ENOS polymorphisms in dizygotic twins. The use of same-gender twins allowed to control for confounders such as age and gender, known to modulate NO production and influence BP. Our data are in favor of an influence of ENOS on BP level in this population." @default.
