- W1978049597 abstract "The level of tryptophan mRNA in several Escherichia coli strains has been quantitatively measured by a procedure based upon competition between labeled and unlabeled RNA molecules for binding sites on φ80 trp DNA. Highly labeled [3H]RNA specifically bound by φ80 trp DNA was isolated from a tryptophan-constitutive E. coli strain. Competition experiments were carried out using this [3H]RNA, φ80pt190 DNA and unlabeled RNA isolated from various E. coli strains. Suitable treatment of the data yielded a numerical measure of the competing power of each RNA preparation. This was insignificantly low for RNA from a bacterial tryptophan deletion mutant, appreciable for RNA from tryptophan-repressed cultures and increased 3.4-fold by tryptophan derepression. Data on the binding stoichiometry between purified Trp-mRNA and φ80trp DNA permit a calculation of the absolute abundance of Trp-mRNA. For a tryptophan constitutive mutant the minimum estimate is 25 polycistronic Trp-mRNA molecules per bacterial cell, as compared to two per cell for wild-type E. coli repressed by tryptophan." @default.
