- W1978387560 abstract "Abstract The circular dichroism spectrum in the visible light region of umecyanin, containing one Cu2+ per molecule, has been studied in detail between pH 7.26 and 11.8. When the spectrum was resolved into Gaussian functions, it was found that umecyanin can form three conformations in the pH interval studied. Conformation Cu2+−1 and Conformation Cu2+−2 can exist in reversible equilibrium around pH 9.5 as a midpoint. Above pH 11.4, umecyanin is transformed irreversibly into conformation Cu2+−3. When the copper is reduced in Conformation 1 and 2, the original spectrum is restored after reoxidation. When the copper is reduced in Conformation Cu2+−3 the Culigand structure is irreversibly lost. Spectrophotometric titration of umecyanin showed that all the four tyrosines are titrated with p K a = 11.7 . It is concluded that tyrosine is of no importance for the transformation between Conformations 1 and 2, but can play a role in the formation of Conformation 3." @default.
