- W1978793802 abstract "Objectives: Integrin molecules have been suggested as markers of endometrial receptivity. We previously reported flow cytometry (FC) to assess the quantitative expression of endometrial integrins during the menstrual cycle (Hum Reprod 1999). In the present study we quantitatively determine the profile of expression of integrins in endometrial stromal (ESC) and epithelial cells (EEC) from fertile women treated with CC for super-ovulation induction and in users of copper-releasing IUD. Design: Comparative study for the quantitative expression of α1, α4, αv and β3 integrins in mid-secretory endometria from fertile women treated with CC, IUD-users and controls. Materials and Methods: All subjects included in this study were fertile with regular and ovulatory menstrual cycles, age ranged from 33–41 years old (mean 36.6). CC-patients: Ten women undergoing laparoscopy for tubal ligation were treated with a daily dose of 50 mg (i.v.) of CC for induction of ovulation. IUD-users: Five women users of T-Cu-380a (Population Council, USA) were included in this study. Fertile controls: Nine fertile women were included in the control group. Tissue samples were taken at the mid-secretory phase or implantation window, 6–10 days after ovulation. The histological dating of endometrial biopsies was assessed according to Noyes’ criteria. Endometrial dating correlated well with the date of ovulation assessed by repeated vaginal ultrasonography. The expression of the integrins α1, α4, αV and β3 was quantitatively determined by FC in dispersions of EEC and ESC isolated from endometrial biopsies using a two-step enzymatic digestion and semi-quantitatively determined (HSCORE) by immunohistochemistry using specific monoclonal antibodies. Results: Forty percent of endometrial biopsies from CC-patients were found as out-of-phase and some cases showed non-synchronized development between stromal and glandular cells. α1, αV and β3 integrin subunits from CC-patients were under-expressed in ESC and β3 in EEC (p<0.05). Differences in β3-ESC and -EEC expression from CC-patients were higher in out-phase-endometria. Endometria from IUD-users showed strong signs of acute inflammation. No differences were observed between IUD-users and controls for the expression of α4, αV and β3-EEC integrin subunits during the implantation window. However, IUD-users over-expressed the α1-EEC and under-expressed α4-ESC (p<0.05). The HSCORE values calculated from the degree of staining of endometria correlated well with those results obtained from FC analysis. Conclusions: CC-treatment for ovulation induction provokes a high frequency of-out-phase endometrium and the disregulation of the expression of integrin markers of endometrial receptivity in fertile patients. Interestingly, β3-integrin was under-expressed in all CC-patients. T-Cu intrauterine device alters endometrial receptivity by a different mechanism independent of the regulation of the most reliable marker of endometrial receptivity, the epithelial β3 integrin." @default.
