- W1978940170 abstract "Rapid flow, isolated perfusion of the kidney has been extended from 5 minutes to several hours, and even to 22 hours in the case of in vitro perfusion. In accomplishing this it was found that the block that occurs 5 minutes after perfusion begins could be eliminated by prior filtration of the perfusate through a glass-wool filter. A second perfusion block occurred 20 to 40 minutes after the onset of perfusion. This block could be eliminated by the addition of a serotonin antagonist, UML-491, to the perfusate. The cause of the first block is most probably platelet aggregates. It is not established whether the second block is caused by further platelet aggregation or by the vasoconstrictor effects of serotonin. A marked decrease in circulating platelets occurred in 5 of the experiments during the first 10 minutes of perfusion, but subsequent changes were either minimal or absent. Kidneys that are perfused for 3 or 4 hours in situ function normally after contralateral nephrectomy. Subsequent examination, however, reveals focal areas of cortical scarring. Reimplantation of kidneys which have been perfused 8 hours has been unsuccessful because a severe clotting defect develops in the recipient dog which results in exsanguination.
