- W1979396362 abstract "The persistence and propagation of mitochondrial DNA was tested in somatic hybrid cell strains derived by Sendai virus-induced fusion of cells of rat and man, and mouse and man. The rodent cells were freshly dissociated from individual tissues of 15-day embryos. Human established lines VA2, and in one ease D98AH2, were used. As described by Coon & Minna (manuscript in preparation) hybrid cells of this kind segregate either human or rodent chromosomes and show considerable stability. The ratio of rodent to human mitochondrial DNA in hybrid colonies was determined by an assay that distinguishes DNAs on the basis of their different nucleotide sequences. This method uses the different hybridization efficiency of complementary RNA to its template and to mitochondrial DNA from the other species. Nineteen rat-human, and seven mouse-human hybrid cell strains were found which contained both parental mitochondrial DNAs when tested approximately 40 to 60 population doublings after fusion. At that time we derived a series of subclones from six of the rat-human strains. Five of these sets of subclones contained both rat and human mitochondrial DNAs in each of the subclones when tested after an additional 25 doublings. This experiment shows clearly that both rat and human mitochondrial DNA molecules have persisted and replicated in individual hybrid cells for at least 40 doublings. Some strains were tested again after continued growth and an additional cloning. Hybrid cell populations containing both rat and human mitochondrial DNAs were found at 150 doublings after fusion. A positive correlation exists between the segregation of chromosomes and of mitochondrial DNA in these hybrid cells.
