- W1979671486 abstract "Photosynthetic carbon metabolism of diatoms is difficult to understand, and experiments conducted by different authors lead to contradictory conclusions. The pattern of carbon fixation was studied in Skeletonema costatum cultured under three different light regimes (continuous light CL; 12 h light/12 h dark cycles: 12 h/12 h; 2 h light/2 h dark cycles: 2 h/2 h). The short-term incorporation of NaH14CO3 (10–60 s) into photosynthetic products showed that 3-phosphoglycerate was the initial product of photosynthetic CO2 fixation. Although this diatom presented a predominant C-3 pathway, 14C incorporation also occurred by β carboxylation (C-4 metabolism). Incorporation into metabolites off β carboxylation was constant whatever the light regime, whereas the importance of the C-3 pathway varied. It is noteworthy that with aging of the culture 14C incorporation decreased and that under continuous light, there was less fixation in Skeletonema costatum by the C-3 pathway. We found that continuous light is not the best light regime for diatoms: C-4 metabolism is enhanced at the expense of C-3 pathway, which is the case under unfavourable conditions. Our results show the importance of light regime for the photosynthetic metabolism of diatoms.Le métabolisme photosynthétique du carbone est encore imparfaitement connu et les expériences menées par différents auteurs conduisent à des conclusions opposées. Le mode de fixation du carbone a été étudié sur des cellules de Skeletonema costatum cultivées sous trois régimes lumineux différents (lumière continue CL; alternances 12 h lumière/12 h obscurité: 12 h/12 h; alternances 2 h lumière/2 h obscurité: 2 h12 h). L'incorporation à court terrne du NaH14CO3 (10 à 60 secondes) montre que le 3-phosphoglycérate est le premier produit formé. Cette diatomée présente donc une voie de fixation de type C-3 préctominante, mais l'incorporation du 14C s'effectue aussi par β carboxylation (métabolisme C-4). L'incorporation dans les métabolites de β carboxylation est constant quel que soit le régime lumineux, alors que la fixation de type C-3 varie. Il est à noter qu'avec le viellissement de la culture, l'incorporation du 14C décroit et qu'en lumière continue, Skeletonema costatum présente une fixation par la voie de Calvin (C-3) moins importante. Nous signalons que la lumiè're continue n'est pas Ie meilleur régime lumineux pour les diatomées: la voie C-4 est favorisée aux dépens de la voie C-3, ce qui est le cas dans des conditions défavorables. Nos résultats montrent donc l'importance du régime lumineux sur le métabolisme photosynthétique des diatomées." @default.
