- W1979766114 abstract "Two main steps of differentiation have been considered for studies of mechanisms controlling hemoglobin synthesis, that is the nucleated erythroid population from the erythropoietic spleen and the reticulocyte population from the circulation of phenylhydrazine-treated mice. The absolute rate of hemoglobin synthesis was measured on a “per cell” basis as pmoles of leucine incorporated into purified hemoglobin per unit of time. By these criteria reticulocytes synthesize hemoglobin at a rate which is 4-fold faster than nucleated erythroid cells. Polypeptide chains growing on polyribosomes become completely labelled with [14C]valine after a 1-min pulse in reticulocytes and after a 2.5-min pulse in nucleated erythroid cells. The specific activity of N-terminal valine in α chains released into cytoplasm becomes equilibrated with the specific activity of internal valine after a shorter period for reticulocytes (about 10 min) than for nucleated erythroid cells (about 20 min). Both lines of evidence suggest that hemoglobin synthesis is faster in reticulocytes because their ribosomes acquire in the course of erythroid differentiation the capacity to translate the hemoglobin messenger at a higher rate." @default.
