- W1979824983 abstract "Spermatogenesis is a complicated process that composed of mitosis of spermatogonial stem cells (SSCs), meiosis and spermiogenesis to form mature spermatozoa. Each stage of spermatogenesis expresses specific markers such as Oct-4, GFR-a, SALL-4, C-kit, CD-9, and others (premeiotic); Crem-1 (meiotic), LDH (meiotic/postmeiotic); Protamine and Acrosin (postmeiotic). Aim: To evaluate the expression levels of premeiotic, meiotic and postmeiotic markers in testicular biopsies from non-obstructive azoospermic patients. The patients (n=14 patients; 16 testes were examined) were defined as azoospermic according to semen analysis. RNA was extracted from testes tissues of the patients and the levels of spermatogenic markers were examined by real time PCR (qPCR) analysis. Our results show that two testes did not express any spermatogenic marker. In all the reminder 14 testes, we have detected premeiotic markers. In 6/14 testes we have detected meiotic markers and in 7/14 testes postmeiotic markers were detected. Also, in 11/14 testes, sperm was identified. In addition, 6/14 testes expressed LDH and sperm was identified in 5/6 of the LDH positive testes. Also, 5/14 testes have expressed protamine, and in all of the 5 testes sperm was identified. In 4/14 testes sperm was identified although meiotic and postmeiotic were not detected. In one testis we identified sperm even though it did not express any spermatogenic marker. In most non obstructive azoospermic patients, one or more premeiotic markers were detected in their testis. Our results emphasize the diversity of non-obstructive azoosermia in relation to type of premeiotic markers expression in their testis. The postmeiotic markers protamine and LDH might be used as good predictors for the presence of sperm in the testis. The RNA expression of premeiotic markers encourages us for possible in vitro maturation of germ cells to sperm, and for future prepubertal male fertility preservation." @default.
