- W1980735090 abstract "Abstract A method for studying the binding capacity of chromatin for actinomycin D, based on the use of tritium-labeled actinomycin and centrifugation through sucrose density gradients is described. In addition, optimal conditions for the assay of the template activity of rat liver chromatin for RNA polymerase are presented, allowing the detection of small increases in the amount of available template. The capacity of rat liver chromatin to bind 3 H-actinomycin D and to support RNA synthesis was about 40 and 10%, respectively, when compared to purified rat liver DNA. These values imply that in native chromatin the binding of RNA polymerase is restricted to a greater extent than is the case for the antibiotic, which, being a smaller molecule, binds to smaller and less specific, unmasked regions of the DNA. The restriction of both parameters is due to the binding of chromosomal proteins to the DNA since extraction of the chromatin with either 0.2 n HCl or increasing concentrations of NaCl results in a progressive increase in the capacity to bind 3 H-actinomycin D and to act as template for RNA synthesis. Incubation of the chromatin with cortisol phosphate for 10 min at 37 ° results in an increase of both its ability to bind the antibiotic (10%) and its capacity to act as template for RNA polymerase (15%). The effect of cortisol phosphate shows a clear dose dependence with optimal concentrations between 10 −6 and 10 −5 , m . This effect of the hormone is probably the consequence of interactions with the chromosomal proteins and does certainly not result from a direct action on either the DNA or the enzyme RNA polymerase. Both effects of the hormone, i.e., on the binding of 3 H-actinomycin D and on the template activity for RNA synthesis, show a certain organ specificity since they could not be reproduced when chromatin from rat thymus or pea seedlings was employed." @default.
