- W1984820901 abstract "A18 Aims: Successful organ transplantation depends on adequate preservation of cellular functions. Cultured hepatocytes have been used to evaluate liver cell injury in preservation. In this study we aimed to establish an in vitro experimental model to evaluate the preserving capacity of various preservation solutions. Methods: To show the suitability of cultured cells in liver preservation studies, a cell line of normal rat hepatocytes was incubated with University of Wisconsin solution (UW), Ringer’s lactate solution (RL), and phosphate-buffered saline (PBS) at 37? and 4 ?, respectively. The cell viability was assessed by propidium iodide uptake and the cell morphology was microscopically documented. Results: At cold temperature UW preserves the hepatocytes without significant cell death for up to 48h, whereas higher than 50% of viability loss was noticed in RL and PBS after 24h incubation. Under warm preservation conditions, cell death was significantly noticed in RL- and UW-treated groups for 6h incubation. The morphological alteration correlated with the positivity of propidium iodide staining. Conclusions: The finding in this study correlates with the clinical practice experience that cold UW solution maintains the integrity of liver organ function for at least 24h of storage time. Cultured hepatocytes may be a useful indicating cell for liver function preservation, and thereby provide a rapid and economic method to initially evaluate the the effectiveness of new formulation in preservation solutions." @default.
