- W1984874037 abstract "The growth of cells is regulated by growth factors which act as ligands of specific receptors on the cell surface. Thus, ligands binding to the receptors activate a series of cellular responses leading to proliferation. In various pathological conditions the receptors can mediate these signals in the absence of ligand, either because the receptors are vastly overexpressed on the cell surface or because the receptors are mutated and thus evade normal regulation of function. The role of the receptor for the epidermal growth factor for the growth and progression of gliomas has long been established. The discovery of additional proteins which share similarities with the receptor for the epidermal growth factor (EGFR) and the identification of more ligands for these receptors initiated a wide spectrum of investigations into their biological relevance. The advent of gene knockout technologies and the discovery of receptor mutants, which can mediate biological effects in the absence of ligands, enabled a deeper insight into the role of these proteins during development, in normal physiology and in disease. It has become apparent that several members of the EGFR family are expressed in the brain and have crucial roles in the brain during development. In addition, some of these proteins are aberrantly expressed in high grade gliomas and other brain tumours, and their expression is intricately involved in the biology of these tumours. This review aims to present an insight into the basic biology of the family of EGFR molecules and their ligands with special emphasis on the brain." @default.
