- W1985051128 abstract "A procedure is described for separation and identification of microquantities of guanidino derivatives found in normal urine with a Dowex 50 ion exchange column using sequential and gradient elution from pH 3.1 to 11.3. The procedure is applied to the urine of normal males and males suffering from chronic glomerular nephritis with nitrogen retention. Arginine, creatinine, guanidinoacetic acid, and creatine are identified in normal urine using alkaline picrate to identify the creatinine, diacetyl reagent to identify the creatine, and the Sakaguchi reaction to identify the arginine and guanidino acid. Arginine is distinguished from guanidinoacetic acid with ninhydrin. In the uremic patients, the guanidinoacetic acid concentration is markedly reduced, and a new Sakaguchi-positive peak appears before creatine on elution at pH 3.1. This substance does not react with diacetyl, alkaline picrate, or ninhydrin. Creatine and creatinine levels are of the same order as that found in the normal individual. The arginine peak is elevated in the uremic patient and two minor Sakaguchi-positive peaks occur at either side of the creatinine peak. An easily assembled apparatus is also described for the gradient elution of the chromatographic column at constant temperature." @default.
