- W1985896272 abstract "Summary The role of nucleotides and phosphate has been studied on the glutamate dehydrogenase activity (GDH) and on preferential oxidation pathway of glutamate in pig heart mitochondria. A comparison has been made with purified GDH from beef liver and pig heart mitochondria. o 1. Substrate and nucleotide contents have been determined in intact pig heart mitochondria. They allow a regulatory role of GDH. 2. A technique is described to study pig heart mitochondria GDH in its environment without permeability effects. 3. Kinetic parameters of GDH in its environment have been determined and compared to those of the purified enzymes (Km Vm, specificity towards NAD, NADP, deamino-NAD and deamino-NADP). 4. Optimal pH is given for NAD or NADP as coenzymes. It agrees with those of purified enzyme. 5. GDH effectors have been studied : as in purified systems, ADP and AMP are activators ; GTP, ITP and GDP are inhibitors. But the heart GDH exhibited a striking Pi sensitivity : Pi enhanced ADP activation and released GTP inhibition. ADP activation increased with pH. 6. In intact mitochondria, all the above parameters cooperated to favour a maximal glutamate oxidation via GDH during state 3 when oxidative phosphorylation operates intensively." @default.
