- W1986373404 abstract "Increased glucose metabolism is one of the most characteristic phenotypes of malignancy. Rat Intestinal Epithelial (RIE-1) cells that contain an isopropyl-1-thio-β-D-galactopyranoside (IPTG) inducible expression system for H-RasVal-12 was utilized to evaluate phenotypic changes associated with oncogenic Ras expression and cellular transformation. We have demonstrated that oncogenic H-Ras expression increases the committed steps of glucose metabolism, namely glucose uptake and phosphorylation in these cells. Induction of H-RasVal-12 expression increases hexokinase 2 (HKII) mRNA 2 - 4 fold by 12 hours. Although hexokinase 1 (HKI) is the predominant isoform in RIE-1 cells, its levels are not altered by H-RasVal-12. Oncogenic H-Ras expression also increases glucose transporter 1 (GLUT1) mRNA and to a smaller extent glucose transporter 3 (GLUT3). Ras oncogenes are known to act through several separate signal transduction pathways including the phosphoinositide 3-kinase (PI3-kinase) and mitogen-activated protein (MAP) kinase pathways. To determine which of these pathways contribute to this mRNA increase, these pathways were separately inhibited in H-RasVal-12 inducible RIE-1 cells and changes in mRNA expression were analyzed by real-time RT-PCR. Inhibition of the PI3-kinase pathway by LY294002 decreases the induction of HKII and GLUT1 in a dose-dependent manner (Figure 1)." @default.
