- W1987818023 abstract "An aqueous extract of antigen was prepared from washed, ruptured trophozoites of Entamoeba histolytica, and dialyzed against distilled water to remove NaCl. It was divided into four aliquot samples, in which the ionic strength of NaCl was adjusted to 0.0, 0.068, 0.136, and 0.272. The adjusted aliquots were fractionated by acid precipitation, and the fractions were assayed for antigenic reactivity in hemagglutination and complement fixation tests, and for content of nitrogen and carbohydrates. The added NaCl exerted a strong influence on the solubility of proteins and antigens on fractionation by acid precipitation. In the absence of added electrolyte, precipitation of hemagglutination antigens was virtually complete at a pH of 4.8, but when NaCl was added to achieve an ionic strength of 0.272, most of the antigen measured by hemagglutination or complement fixation tests was soluble. The concentration of nitrogen in the fractions paralleled the reactivity of antigen when assayed by the hemagglutination test, but not the complement fixation test. The activity of complement fixing antigen more closely paralleled the carbohydrate content of fractions. The significance of these findings was discussed in relation to reproducibility of fractionation studies and the composition of amebic antigens." @default.
