- W1988156767 abstract "Vaccine approaches against AIDS have focused on inducing cellular immune responses, since many studies revealed the role of T cell responses in the control of human immunodeficiency virus or simian immunodeficiency virus (SIV) infections. The experimental infection of rhesus macaques with SIV or chimeric SHIV is routinely used as a model for AIDS. In such models, DNA immunization is a tool to elicit specific T cell responses and to study their protective efficacy. DNA immunogenicity in primates depends on parameters such as level of antigen expression, choice of the antigen among SIV proteins, use of fusion proteins, route of immunization, and addition of adjuvants. Recent results suggest that priming with DNA and boosting with attenuated recombinant viral vectors, each expressing corresponding SIV antigens, leads to improved specific immunity and, in some cases, affords protection against pathogenic challenge. After preclinical evaluations, DNA has entered clinical trials for a therapeutic or prophylactic gene-based AIDS vaccine. Les nouvelles approches vaccinales contre le SIDA visent principalement à induire une réponse immunitaire cellulaire. En effet, de nombreuses études ont montré le rôle des lymphocytes T dans le contrôle de l'infection par les virus de l'immunodéficience humaine (VIH) et simienne (VIS). L'infection expérimentale du macaque rhesus par le virus VIS ou par un virus chimérique VIH/VIS est couramment utilisée comme modèle du SIDA humain. L'immunisation à base d'ADN ou vaccination génétique apparaı̂t comme un outil de choix pour l'induction de réponses T dans ces modèles. Ce nouveau mode d'immunisation a été beaucoup amélioré au cours des dix dernières années. Plus récemment, de nombreuses études suggèrent que la combinaison entre vaccins à base d'ADN et vecteurs viraux atténués exprimant des antigènes communs permet d'améliorer de manière significative les réponses immunitaires spécifiques. Après de nombreuses évaluations précliniques chez l'animal, ces vaccins, aussi bien prophylactiques que thérapeutiques, sont entrés dans des phases d'essais cliniques chez l'homme." @default.
