- W1988318604 abstract "Post-genomic medicine has the potential of generating pathways which allow better comprehension of varicocele-associated male infertility and generate potential biomarkers for diagnosis and treatment. Thus, we set out to determine lipid and protein pathways in seminal plasma (SP) of adolescents before and after varicocelectomy. Prospective study. 15 adolescents participated in the study. SP was frozen before and 3 months after subinguinal microsurgical varicocelectomy. Patients were grouped as (i) pre and post-surgery (paired Student's T-test, P<0.05), and (ii) as those presenting at least 10% improvement in semen quality after surgery (Mann-Whitney test, P<0.05). Lipids were extracted from SP and identified using MALDI ionization, and the online databases Lipid Maps and KEGG. SP proteins were included from a previous study for pathway generation. Post-genomic pathways were generated and functional enrichment of Gene Ontology (GO) terms was analyzed (P<0.01). LASSO algorithms were used to search for ligand activity in differentially present lipids. Regarding LASSO activity 4 lipids were more intense in the pre-surgery samples related to glucocorticoid and peroxisome proliferator receptor activity, and 2 in the post-surgery samples with metalloenzyme activity. Also, 5 lipids were associated to improved outcome at 3 months post-surgery, with mainly androgen receptor and retinoic acid receptor-binding activity. Protein and lipid pathway maps demonstrated enriched Phospholipase A2, retinoic acid, and oxidoreductase molecular functions, and arachidonic acid catabolism, hydrogen peroxide catabolism, and negative regulation of protein glycosylation biological processes, under GO analysis. Varicocelectomy in adolescents leads to altered SP lipid fingerprints. Lipid profiles may be used to predict outcome in these adolescents. Observation of the post-genomic pathways may assist comprehension of mechanisms in male infertility." @default.
