- W1989480195 abstract "Improving pregnancy rates associated with the use of cryopreserved human oocytes would be an important advance in human assisted reproductive technology (ART). The objectives of this study are to evaluate the survival rates and pregnancies of cryopreserved human oocytes following vitrification. Retrospective evaluation of oocyte survival rates post-thawing and pregnancy rates following transfer the resulted embryos. The Hospital Research Ethics Board (REB) has approved the protocol. The patients were given fully explanation for the procedures and a written consent forms were obtained. A total of 15 women were included in this report until submission, in which women undergoing ovarian stimulation for intrauterine insemination (IUI) had their mature and immature oocytes collected and vitrified before cancellation the treatment cycle due to the high risk of multi-fetal pregnancy. The mean age of women was 31.7±3.7 years. The mature oocytes were subjected to vitrification immediately after collection, and the immature oocytes were cultured in IVM-Medium for maturation, and all in vitro matured oocytes were vitrified. The vitrified oocytes were thawed and inseminated using intracytiplasmic sperm injection (ICSI), and the resulting embryos were transferred in a subsequent menstrual cycle. As the procedure of vitrification, the oocytes were suspended in equilibration solution (7.5% ethylene glycol and 1,2-propanediol) for 5 min, and then the oocytes were transferred to vitrification solution (15% ethylene glycol and 1.2-propanediol +0.5 M sucrose) for 45-60 seconds at room temperature. The oocytes were loaded on a special designed device, Cryoleaf, and immediately plunged it into liquid nitrogen for storage. For thawing, the Cryoleaf was directly inserted into 37°C thawing solution (1.0 M sucrose) for 1 min. The thawed oocytes were transferred to 0.5 M and 0.25 M sucrose solutions for 3 min respectively, and then washed twice with culture medium for insemination. A total of 180 oocytes were vitrified and thawed from these 15 women. Of those, 169 (93.9%) oocytes were survived post-thaw. Following insemination by ICSI, 126 oocytes were fertilized normally (74.6%), and 7 patients became pregnant (46.7%) after transferring the resulted embryos (3.6±1.3 embryos per patient). Implantation rate was 20.4% (11/54) This preliminary results indicate that human oocytes can be cryopreserved successfully with the procedure of vitrification. This vitrification method may be efficiently used for cryopreservation of human oocytes." @default.
