Body weight was determined daily. At the end of experiments, colonoscopy was performed and colon samples were obtained for H&E staining. Results: The deletion of PTPN2 in intestinal epithelial cells had only minor impact on the course of intestinal inflammation: PTPN2xVilCre mice showed only a trend to earlier onset of weight-loss and a significantly increased murine endoscopic index of colitis severity (MEICS) but no difference in histological severity of inflammation when acute colitis was induced by 2.5% DSS. When acute colitis was induced using 2% DSS, no significant differences were observed. In chronic DSS colitis PTPN2xVilCre mice showed a trend towards less severe inflammation compared to control littermates with significantly decreased MEICS. However, we noted pre-stage adenoma-like epithelial transformations in most DSS treated PTPN2xVilcre mice. Discussion: Our data demonstrate that, in contrast to T-cell specific loss of PTPN2, PTPN2 deficiency in IEC has only minor impact on inflammation in DSS colitis. The role for dysplasia formation on the other hand is well in line with the role of PTPN2 in epidermal growth factor (EGF) -receptor signalling. The role of PTPN2 as a potential tumor suppressor in IEC warrants further investigation" @default.
