In chimeric mice constructed by a graft of one parental strain thymic tissue on athymic F1 (C57BL/6 × BALB/c) mice, the F1 T cells that mature provide helper activity for B cells that are H-2 compatible with the graft donor. A T cell growth factor (TCGF)† has been characterized that has helper T cell-replacing properties in the induction of in vitro immune responses. TCGF appears to exert its effects via the clonal expansion of antigen-activated T cells present in these cultures. The T cell-growth promoting properties of TCGF have been used to establish continuous cultures of proliferating helper T cells. Thus, by utilizing F1 thymus-chimeric mice to select for cellular recognition specificities in T cells followed by antigen-activation of T cells to render them responsive to TCGF, we hope to generate clones of helper T cells from a common genetic background to analyse the biochemical nature of their recognition structures." @default.
