- W1991775253 abstract "Exponentially growing cells or penicillin protoplasts of Escherichia coli were allowed to incorporate 32P briefly. Cells, lysozyme lysates of cells, and osmotic lysates of penicillin protoplasts were subjected to sonic oscillation, followed by centrifugal fractionation and extraction of nucleic acids. 1. DNA of untreated lysates was largely in the particulate fractions sedimenting at low centrifugal forces. Brief sonic treatment shifted most of this material into the high-speed supernatant. However, a small amount of DNA was not solubilized even after extensive sonic treatment, and this refractory material was very rich in newly-synthesized DNA. 2. In lysates of whole cells, sonic oscillation did not alter the gross distribution of RNA between centrifugal fractions, but it did preferentially solubilize a very small amount of newly-synthesized RNA. In lysates of penicillin protoplasts, sonic oscillation solubilized much of the ribosomal RNA, but in this case the material shifted out of the particulate fractions had only the average content of newly-synthesized RNA. 3. A possible interpretation of our findings is that the bacterial chromosome, like that of higher plants, replicates sequentially. According to this view, DNA synthesis is continuous only in the sense that some of a cell's DNA is replicating at any instant, not that all DNA is continuously involved in the replication process. Some of our results suggest that when a given portion of the genome is not replicating it may be participating in RNA synthesis." @default.
